Stability has more than one endpoint
Lyophilization changes water activity and can improve handling, but the result depends on peptide sequence, excipient, cake structure, container closure, residual moisture, reconstitution, and storage environment. A stability claim should identify which endpoint was measured: intact assay, related substances, aggregation, appearance, reconstitution, or biological function.
A measured formulation example
In a 2017 CSP7 study, solution retained more than 95% peptide at 5 and 25 °C for up to 48 hours and tolerated at least five freeze-thaw cycles. Lyophilized cakes with excipients remained above 96% for up to four weeks at room temperature, and more than 98% remained in solution eight hours after reconstitution at room temperature. Protected from moisture at 5 °C, formulations were stable for up to 10 months; exposure to 75% relative humidity increased absorbed moisture and induced reversible aggregates. The study used a specific peptide and formulation set, so these percentages are not generic specifications.
